As @maksimov noted in the comments, a 64 bit JVM cannot load the 32 bit Notes DLLs. There is, howver, a 64 bit version of the DLLs, but you will have to install and configure the Lotus Domino Server (64 bit Windows version) on your machine - and on any machine(s) where the web application will run. You will only have to run the Domino server once per machine in order to get it fully configured. It will not have to be running all the time in order for your code to access the DLLs, but you will have to leave it installed. You will have to, however, check with whomever manages your IBM relationship, and possibly with IBM, in order to determine whether there are any licensing and cost implications before installing the Domino server code this way. (One further note: in order to get the 64 bit Domino Server code cleanly installed on the machine and insure that that particular version of nlsxbe.dll is the one that your code loads, you will be best off if you uninstall and completely clean the existing 32 bit Notes client installation. And by cleaning, I'd get rid of the lotus.ini file, and any notes.ini files and Notes-related registry entries that might be left over after the uninstallation.)

The Lotus Domino connector uses the Lotus Notes client to communicate with Lotus Domino Server. As a consequence of this dependency, a supported Lotus Notes Client must be installed on the synchronization server. The communication between the client and the server is implemented through the Lotus Notes .NET Interop (Interop.domino.dll) interface. This interface facilitates the communication between the Microsoft.NET platform and Lotus Notes client and supports access to Lotus Domino documents and views. For delta import, it is also possible that the C++ native interface is used (depending on the selected delta import method).
Create, Update, and Delete ResourcesThe Create, Update, and Delete operations are performed by the Lotus Domino connector in the Resource Reservation database. When a database is designed to receive a mail, a Mail-In Database document is created in Lotus Domino. This document must exist in Domino Directory of every server that stores a copy of the database. The Domino Connector relies on the feature Directory Assistance to be able to find the secondary address books. If the secondary address books are missing, verify if Directory Assistance, previously at _8.5.3/com.ibm.help.domino.admin85.doc/H_ABOUT_DIRECTORY_ASSISTANCE.html has been enabled and configured on the Domino Server.
Behavior change: You no longer have to install client-side, browser-specific components for the web client. The applet now downloads automatically if you don't already have the applet version that matches the server.
